Research Topic:
Most cell death in mammals is executed via the `intrinsic` or `mitochondrial` apoptosis pathway regulated by the Bcl-2 family. It has been anticipated for a long time that Bcl-2 proteins can substitute for each other in inhibiting cell death in response to genotoxic stress and that overexpression of any given BH3-only protein will kill a cell, while overexpression of Bcl-2 or related Bcl-xL or Mcl-1 blocks cell death, in a rather non-specific manner. However, recent biochemical and genetic evidence suggests that BH3-only proteins proteins act frequently in a non-redundant manner to neutralize the pro-survival function of selected Bcl-2-like pro-survival molecules expressed in a given cell type. Hence, parts of our laboratory investigate the role of selected Bcl-2 family proteins in cell death in the immune system employing mouse genetics.
Next to Bcl-2-controlled cell death, alternative cell death signalling pathways have been described, among which, the so-called PIDDosome and caspase-2 regulated cell death pathway caught our attention. We hypothesize that caspase-2 is critical for cell death in response to cell cycle check-point failure and try to understand within the MCBO program how caspase-2 is activated under these conditions and triggers cell death in p53-defective tumor cells.
Finally, out of these studies mentioned above, we developed an interest in the role of autophagy in cell death responses and plan to explore the role of an ill-defined protein, PDCD5, in this process using mouse genetics.
