Persistent infections such as HIV, hepatitis B and C virus are major causes of mortality world-wide. Dendritic cells (DCs), the sentinels of the immune system, are essential for the generation of a protective antiviral immune response. Nevertheless, they are exploited by the viruses to evade innate and adaptive immunity. Upon entering the body via mucosal surfaces, HIV-1 is spontaneously opsonized with complement fragments due to activating the classical complement pathway even in the absence of specific antibodies. After seroconversion, the virus is additionally coated with specific antibodies. Studies involving the differential opsonization pattern of pathogens are an important issue in scientific research, which is rarely addressed in the literature. Due to addressing various receptors, the opsonization pattern of pathogens has a strong impact on the antigen-presenting capacity of DCs. In our group, we are studying different antigen-presenting cells exposed to opsonized/non-opsonized pathogens and consequently the impact of the opsonization pattern on specific cell functions. Therefore, to mimic the in vivo situation more closely, we are integrating innate (complement) and adaptive (antibodies) humoral immune components in all our experimental set-ups.
